The TNF-α, CXCL-8, Ang II, and Ang-(17) concentrations in the cell culture supernatant were calculated via ELISA (Shanghai Hengyuan Biotechnology Co, Shanghai, China). ELISA test was conducted according to the manufacturer’s instructions. Briefly, the standard (50 μL) and the sample were tested (50 μL) by reacting at 37 ?C for 30 min, and the plate was washed 5 times. Afterward, the enzyme-labeled antibody (50 μL) was added to react at 37 ?C for 30 min, and the plate was washed 5 times. Then added chromogenic solutions (A (50 μL) and B (50 μL)) and incubated at 37 C for 15 min. At the end added the termination solution (50 μL) and measured the OD value at 450 nm immediately. The concentration of TNF-α, Ang II, and Ang-(1? 7) was expressed as ng/L. The concentration of CXCL-8 was expressed as pg/mL.
